In this work, we prepared [PtACHTUNGTRENUNG(C^N^N)Cl] complexes 1–3
(Figure 1) containing modified 6’-phenyl-2,2’-bipyridine ligands,
and found these complexes to have the following
properties: i) 3MLCT emission in the visible spectral region
(the long emission lifetime of phosphorescence can enhance
image signal stability and reduce the background noise arising
from fluorescence), ii) solubility and stability in aqueous
staining solutions at room temperature (thus the use of organic
solvents is not required for the optimal sensing of proteins),
and iii) binding to proteins via non-covalent interaction.
By employing these complexes, we have successfully
demonstrated the practical application of luminescent platinum(
II) complexes for protein staining and live cell imaging.
Common spectrophotometric methods, such as the Brad-
ACHTUNGTRENUNGford,[10a] bicinchoninic acid (BCA),[10b] and Lowry assays,[10c]
which are used for protein quantification, in general are
hampered by the interfering agents found in the sample
preparation buffers, require relatively large sample volumes,
have limited dynamic range, and exhibit responses which
are affected by the structure of proteins, the amino acid sequence,
and the isoelectric point (pI). Thus the developmentof analytical tools to detect proteins with high sensitivity,
good binding linearity, small protein-to-protein variation,
and low consumption of samples is of particular importance
in biochemistry.
